Abstract:
To analyze the growth dynamics of different categories of gestational trophoblastic diseases with respect to their levels of cell proliferation and cell death. The proliferation and apoptotic markers in the various categories of GTD will be compared with each other and with the normal placenta to evaluate the role of growth dynamics in the pathogenesis of these diseases. Correlation with clinical outcome of the patients will be made to assess the prognostic significance of the markers. Possible effects from relevant oncogenes and growth factors will also be elucidated.

To determine which members of the newly discovered gene family of angiopoietins are expressed in human tumours by Reverse Transcription-Polymerase Chain Reaction (RT-PCR); to identify the cellulr sources of angiopoietins in tumours by In Situ Hybridisation (ISH); to study the expression of Tie2 in tumours by RT-PCR and ISH; to compare the expression patterns of angiopoietins and Tie2, and determine the role of these molecules in tumour angiogenesis.

Abstract:
To produce a monoclonal antibody that is able to distinguish between the wild-type LMP1 and the deleted (CAO) form; to test suitable antibodies on cell lines and tissue samples to determine if this antibody can distinguish between the wild-type and deleted forms on archival tissue; to analyze by PCR the C-terminal latent membrane protein 1 (LMP1) sequence in HK Chinese nasopharyngeal carcinomas and determine if they contain a 30 bp deletion or mutations that are different from the prototype B95-8 strain; to compare the reaction of the antibodies with the sequence in the NPC samples analyzed by PCR to identify the epitope recognized by the antibody.

Abstract:
1. To determine whether NL escape the immune recognition by down regulating the expression of MHC-I, MHC-II or transporter protein associated with antigen presentation-1 (TAP-1). Many viruses have evolved strategies to interfere with the process of MHC/viral peptide presentation to CTLs(called "stealth strategy"). 2. To determine whether the activated CTLs in the vicinity of NL tumor cells are inhibited by the certain immunoregulatory cytokines produced locally by either the tumor cell themselves or by the reactive influence of cytokines secreted locally, the NL tumor cells are able to proliferate by either autocrine or paracrine production of cytokines. 3. To determine whether the membrane bound Fas-L is constitutively expressed on the tumor cells of NL, and whether Fas expression is down regulated in these tumors since the Fas-L/Fas is one of the major effector systems of CTLs. The levels of Fas-L will also be determined in the sera of the patients with NL. When Fas-L expressed on the surface of tumor cells is cleaved, the soluble form of sFas-L may be released into the circulation and may cause the multi-organ failure often seen in NL patients.

Abstract:
To identify and characterize, by employing 5' RACE technique, the heterologous promoters fused to the BCL-6 gene which deregulate the expression of BCL-6 oncogene in gastric high-grade lymphoma of MALT-type in a larger series of cases with rearranged BCL-6 genes; to study the pattern of expression of the BCL-6 partner genes by Northern blot hybridization analysis, at various stages of B-lineage differentiation and in B-cell lymphoma.
