Abstract:
The project aims at demonstrating the following hypotheses: 1) repeated inflammation may contribute to cancer formation in the colon; 2) COX-2 and 12-LOX could play a co-regulatory role in the pathogenesis of colorectal cancer (CRC); and 3) cigarette smoking could modulate the expression of these enzymes and increase CRC carcinogenesis.

Abstract:
To identify the signal transduction pathways of the adenosine-induced hyperpolarization in the coronary arterial smooth muscle cells and endothelial cells. We hypothesize that the adensoine-induced hyperpolarization in vascular smooth muscle cells and endothelial cells is mediated via cAMP-independent pathways and involves more than one K+ channels; to identify the mechanism of regulation of extracellular adenosine level in the coronary arterial smooth muscle cells and endothelial cells.

To evaluate the effect of cholestin on cholesterol metabolism in human hepatoma cells (HepG2) and to compare its effect with known lipid-lowering agents i.e. statins; to elucidate the biochemical mechanisms of cholestin on lipid metabolism.

To study the acute vascular effects of 17b-estradiol in male as well as female rats. In addition, the modulatory actions of long-term treatment of 17b-estradiol on arterial relaxation will be examined in ovariectomized or sham-operated rats. To examine five hypotheses: a) the lack of NO favors endothelium-dependent contractions: b) the endothelium of diabetic arteries releases more vasoconstrictor breakdown products of arachidonic acid: c) endothelium-dependent contractions are due to a diffusible factor (EDCF); d) oxygen-free radicals play a key role in EDCF-mediated responses: and e) high glucose mimicks the impact of diabetes on endothelial function. These studies should yield a better understanding of the mechanisms(s) underlying diabetic endothelial dysfunction, and hence help to improve the prevention and/or therapy of one of the most feared complications of the disease.
