Abstract:
To genetically manipulate carotenoid biosynthesis in the green microalga chlamydomonas reinhardtii so as to produce the red ketocarotenoid astaxanthin; to develop an optimal growth system for high-yield production of astaxanthin by the engineered organism; to construct plasmids containing single crtO, crtZ or psy genes under the control of a synthetic inducible promoter and construct a bi-directional polar promoter for directing the expression of crtO, crtZ and psy; to introduce the gene crtO and/or crtZ coupled with psy into C. reinhardtii; to investigate the expression of transgenes in C. reinhardtii; to investigate growth kinetics and productivity of astaxanthin of the C. reinhardtii mutants under various culture conditions.

Abstract:
The project attempts to localize BjHMGS by using: 1) anti-BjHMGS antibodies in western blot analysis and immunoelectron microscopy; and 2) BjHMGS-Green Fluorescent Protein fusions. The research team will also identify the amino acid residues in BhHMGS that are essential for catalytic activity by site-directed in vitro mutagenesis. This would be a prerequisite to the crystallization of BjHMGS and the elucidation of its 3-D structure and interaction with substrates, inhibitors and other factors affecting its biological activity.

To elucidate the function of a putative acyl-CoA binding protein, ACBP3, in Arabidopsis thaliana that my laboratory has identified from the Arabidopsis genome sequence; to establish that ACBP3 binds acyl-CoA esters, thus proving it is an acyl-CoA binding protein; to study the pattern of ACBP3 expression in order to elucidate its role in the plant.

Abstract:
The cereal industry in this region is incredibly diverse. There is a vast expansion of diversity fueled by increasing consumer affluence in China, and by the need for convenience products. What attributes of the cereal grain raw materials are most relaated to quality of these products? What additives and modified starches will be in most demand in future? Without basic scientific analysis of products on the market, these questions cannot be answered.

Abstract:
In this proposal, we will construct vectors to produce dsRNA signals in transgenic plants in order to test if mammalian genes in mice and rats can be suppressed by this approach. This work will help set the stage to test if transgenic plants can be used deliver dsRNA signals to suppress expression of specific target genes in mammals. If successful, this would open a novel approach for gene therapy of certain diseases as well as systemic functional studies of all mammalian genes.
