To study the distribution of rat secretin and secretin receptor in various regions of the rat brain by Reverse transcript-polymerase chain reaction (RT-PCR), Northern blot analysis, in situ hybridization and immunocytochemical staining; to investigate the electrophysiological functions of secretin in regulating purkinje cell functions, particularly on neuronal pathways and mechanisms.

Abstract:
To identify target genes that are involved in junction disassembly between testicular cells by an in vitro cadmium chloride disassembly model system. Peptides of 20-25 amino acids will be synthesized based on the extracellular domains of Cx-33 (a testis-specific GJ protein) and occludin (a TJ protein found in the testis). 2.a. These peptides and their corresponding antibodies will be used to perturb cell junction assembly in vitro. 2.b. The active peptide will be injested intratesticularly to assess its effects in vivo. To use antisense oligodeoxynucleotides (ODNs) specific to Cx33, a testi-specific GJ protein largely expressed by germ cells, to perturb the GJs between testicular cells. It is anticipated such a disruption will impair germ cell movement when communications between Sertoli and germ cells are disrupted. 3.a. Antisense ODNs will be screened in vitro by using Sertoli cell cultures and Sertoli-germ cell cocultures to select ODNs that can arrest the timely expression of Cx33. 3.b. Selected ODNs will be injected intratesticularly to assess their effects in the seminiferous epithelium in vivo.
