Abstract:
Avian influenza viruses can directly impact on human and animal health and thus cause economic loss. The study aims to establishing a reverse genetic system and to determine the biological significance of the deletion of NA stalk and the NS genes, particularly in relation to the host-restriction and pathogenesis. Results can enhance understanding of the machanism of inter-species transmission of influenza virus.

To observe the epidemiological dynamics of influenza viruses between domestic ducks (the major influenza gene reservoirs in this region) and migratory birds in southern China; to understand the mechanisms of inter-species transmission of influenza virus from wild aquatic birds to domestic poultry, or vice versa; to determine whether migratory birds may contribute some gene segments for the generation of influenza viruses of pandemic potential.

Abstract:
To conduct an effective, individual EBV antigen expression system which will allow us to obtain large amount of such antigens; to detect if the DCs from NPC patients are selectively functionally defective to individual EBV antigens, and if so, whether cytokine-activate DCs may overcome the deficiency; to ascertain whether the cytokine-activated DCs of NPC patients plus the antigens can ex vivo activate specific CTL precursors or stimulate naive T cells to become specific CTLs, including optimization of the conditions for expansion of these specific (CTL cell lines in vitro; to evaluate whether the activated DCs together with the EBV antigens may indeed stimulate the target specific CTLs in NPC patients after removal of the cancer by traditional therapy.

Abstract:
The vitality of the map gene in Gram positive bacteria, such as staphylococcus, streptococcus, and bacillus, however, is still elusive. Demonstration experimentally of the essentiality of map gene in Gram positive bacteria will subsequently validate methionine aminopeptiase as an effective antimicrobial target against a variety of pathogens, from fungi to bacteria.

Abstract:
In conjunction with conventional genetic approaches (such as gene cloning and gene inactivation, etc.), we set out to employ chemical genetic approach - the use of high-throughput screening (HTS) technologies to identify biologically active small molecules that will interfere with particular cellular processes/biological pathways/gene products - to dissect the molecular basis of virulence and dimorphism in Penicillium marneffei. Forward chemical genetics will be used to identify small molecule compounds that will interfere with host-pathogen interactions and dimorphism in P. marneffei. Reverse chemical genetics will be used to identify small molecules that will have specific inhibitory effects on MetAP1 or MetAP2 (essential enzymes involved in removal of N-terminal methionine) of P. marneffei. Resulting specific inhibitors of MetAPs will be tested in in vitro intracellular survival and in vivo animal infection models.

Abstract:
This project is a multi-institutional collaborative study for developing a high throughput DNA array to allow genotyping of influenza virus isolates of avian origin from Hong Kong and applying it to characterize the influenza virus gene pool in sourthern China. Through its application, it is expected to:- 1) identify the precursor virus that donated its internal genes to generate the mutiple reassortment of H5N1 isolated in Hong Kong markets in 2001; 2) define the frequency of reassortment between viruses of different virus lineages that co-circulate in poultry in Hong Kong and correlate this to host-specificity; and 3) investigate whether avian internal gene segments are transiently introduced into the human influenza gene pool.

Abstract:
To refine existing PCR-based assays for SARS-CoV detection; to develop novel quantitative PCR-based assays which target at other regions in the SARS-CoV genome; to identify the optimal clinical samples for SARS-CoV diagnosis; to determine the quantity of various viral RNA species (i.e. genomic, subgenomic RNA and defective interference viral particle) in clinical samples; to apply the quantitative PCR assays to determine the viral dynamic in SARS patients at different time points; to quantitative the amount of SARS-CoV in atypical SARS patients, such as asymptomatic patients and 'super-spreaders'.

Abstract:
The recent use of high-throughput screening (HTS) to search for small molecule inhibitors provides a new possibility of identifying small molecules with anti-influenza effects. This approach has been proved to be useful in finding small molecule inhibitors of angiogenin. We intend to apply HTS technology to identify small molecules to inhibit influenza virus replication. In particular, we intend to identify inhibitors that are specific for the viral polymerase subunits.

Abstract:
To answer the following questions: (1) the number and sizes of the chromosomes (karyotyping) of P. marneffei; (2) the phylogenetic relationship between P. marneffei and other fungi; specially, whether a sexual stage of it exists in nature which explains why it is difficult to isolate the fungus from environmental samples; (3) to perform a random exploration of the P. marneffei genome and analyze 2000 random sequence tags of the genome of the fungus. This would enable us to better understand the basic mechanisms of dimorphism, biochemical properties, disease pathogenesis, virulence, and immune defence and thus pave the way to treat or prevent the disease.

To characterize 48 S. bovis strains isolated from the blood cultures of 37 bacteraemic patients over a six-year period using a combination of phenotypic and genotypic techniques. The epidemiology, underlying diseases, clinical disease associations, and outcome of S. bovis bacteraemia in relation to the different biotypes and genotypes of S. bovis will be investigated, and the molecular epidemiology of erythromycin resistance in the S. bovis strains will also be analyzed.

To search for a novel antigen of which the immune response against it is life long, such antigen would be crucial in the understanding of the seroepidemiology of the infection, as well as how the pathogen is transmitted exactly. Furthermore, such an antigen may be important in the development of vaccine in penicilliosis marneffei. This could be important to reduce the need for secondary prophylaxis as these patients may have to take antifungal for life.

Abstract:
To test the hypothesis that some of amino acid residues in spike (S) and matrix (M) proteins that are different between human SCoV and animal SCoV-like viruses might determine whether the virus infects human or animals and that peptides designed based on these substitutions might interfere with the formation of fusion-active structures and therefore block the entry of the virus into host cells.
